Lysozyme susceptibility assay. A) Lysozyme treatment growth curves for M. smegmatis/Rv1096 and wild-type M. smegmatis. M. smegmatis/Rv1096 (square) and wild-type M. smegmatis (triangle) were grown in LBT medium at 37°C to an OD600 of 0.2; the cultures were then divided into two parts. One part (closed symbol) was treated with lysozyme, the other part was not. Three microliter samples from each culture were collected at 1 h intervals for OD600 measurements. M. smegmatis/Rv1096 showed significantly greater resistance to lysozyme than did wild-type M. smegmatis (**P < 0.01). Values are means ± SD.
