B. pseudomallei wildtype but not the T3SS3 mutant induces p65, IκBα and TAK1 phosphorylation. A) HEK293T cells were infected with B. pseudomallei strains at MOI 50:1. Cells were lysed at 2 and 3 hr and analyzed by Western blot with anti-phospho-p65, anti-p65 and anti-β-actin antibodies. B) HEK293T cells were infected with B. pseudomallei and B. thailandensis strains at MOI 50:1. Cells were lysed at 2 hr and analyzed by Western blot with anti-phospho-IκBα and anti-IκBα antibodies. C) HEK293T cells infected with KHW at MOI 50:1. Cells were lysed at 1, 2 and 3 hr. Lysates were immunoprecipitated with anti-TAK1 antibody and immunoblotted with phospho-TAK1 antibody. The TNFα stimulated cells were used as a positive control. D) HEK293T cells were infected with B. pseudomallei and B. thailandensis strains at MOI 50:1. Cells were lysed at 2 hr.
