Class I PI 3-kinase p85α-p110 is required for Rck- and PagN-mediated internalization. CHO (a-b) or Jeg-3 (c-d) cells were incubated with AS604850 at the indicated concentrations for 2 h 30 prior to the addition of HB101-pagN (a-b) or MC1061-rck (c-d) at MOI 1:10 for 1 h at 37 °C. Percentages of total cell-associated bacteria (a-c: grey bars) and internalized bacteria (b-d: white bars) were determined as described in the Materials and Methods. Results acquired with drugs are expressed relative to values acquired for the same amount of DMSO-containing medium (DMSO), set at 100%. CHO (e-f) and Jeg-3 (g-h) cells transfected with ∆p85α and Wp85α were infected with HB101-pagN (e-f) or MC1061-rck (g-h) at MOI 1:10 at 37 °C for 1 h. The percentages of total cell-associated bacteria (e-g: grey bars) and internalized bacteria (f-h: white bars) were calculated and expressed relative to values obtained for Wp85 α transfected cells, set at 100%. Values represent means ± SD of three independent experiments with two infected wells per experiment. Results were compared using a Mann Whitney test (***p < 0.001, **p < 0.01)
