Mass spectrometry-based proteomics workflow of K. pneumoniae. Bottom-up proteomics workflow for profiling the cellular proteome and secretome of K. pneumoniae under zinc-limited (M9 minimal media with 0 μM zinc) and zinc replete (M9 minimal media supplemented with 10 μM zinc) conditions. Proteins were extracted from cells and supernatant followed by enzymatic digestion and purification on C18 resin tips. The purified peptides are measured in the first MS scan and peptide fragmentation patterns are observed in the second MS scan (MS/MS).
