YpeB-Cys can be cross-linked in spores. Ten optical density units of decoated dormant spores were incubated with 5 mM APB for 30 min at 37 °C in reduced light and then irradiated with UV light for 15 min. Cross-linked spores were lyophilized and mechanically broken, and proteins were extracted. Whole spore lysates were then visualized via Western blot using anti-YpeB antibodies [20]. The positions of protein size markers are indicated on the left. The YpeB monomer is indicated by the arrow at 50 kDa, higher migrating bands are indicated by arrows at ~ 100 and 150 kDa in YpeB-Cys cross-linked samples
