HrcU yeast two-hybrid interaction assays (Y2H). Native HrcU, HrcUN266A and C-terminal (CT) truncations of both proteins were cloned into the bait vector pGBKT7. HrpJ and HrpP were expressed from the prey vector pGADT7. Blue coloration indicates the strength of protein–protein interactions. All Y2H interactions were quantified relative to an empty-vector control using ImageJ software and tested for significance using Kendall rank correlation coefficient τ tests. Both full-length and truncated HrcUN266A exhibited impaired interactions with HrpP and HrpJ relative to native HrcU constructs
