Inhibition of CD1d impedes SEB binding to RPTECs: 1 × 10 6 cells/ml RPTECs were treated with mouse anti-CD1d antibody (1μl/ml) or PBS for 30 m; post-wash, the cells were exposed to 100 μg/ml SEB for 1 h. Anti-SEB rabbit antibody followed by goat anti-rabbit FITC-conjugated secondary antibody was used for reporting purposes. A “no cell” control was used as baseline and the average was used to normalize intensity (Treated – Control ± SE) (N =5); Welch’s t-test p value cut-offs were designated by * < 0.05. The first bar (designated as SEB + CD1d) shows SEB-CD1d binding in the absence of the inhibitory anti-CD1d antibody. A significant reduction of signal was observed in presence of the inhibitory anti-CD1d antibody (second bar; designated as SEB + inhCD1d). Third bar plot (inhCD1d) shows the specificity of the anti-SEB primary antibody and the corresponding secondary antibody towards the anti-CD1d antibody in the absence of SEB exposure.
