The bacteria were grown at 25°C in LB medium, suspended in minimal M9 medium and preincubated for four hours either at 25°C (A) or at 37°C (B). After 60 min at 37°C, no significant AK activity was detectable, while it remained high at 25°C (not shown). Then glucose was added at 25 and 37°C (zero time) and aliquots were taken after various time intervals for the determination of thiamine derivatives. The results are expressed as mean ± SD (n = 3).
