SiRNA silencing of IL-10 enhances C. butyricum -induced NF-κB activation and IL-8 secretion. HT-29 cells were transfected with siNEG (negative control-specific siRNA) or IL-10-specific siRNA for 48 h prior to stimulation. RNA was extracted after a 2 h C. butyricum treatment, and the levels of IL-10 (A) and IL-8 (B) were measured by real-time PCR. (C) IL-8 secretion in response to C. butyricum in siNEG control and IL-10 knockdown cells. (D) Immunoblot shows levels of NF-κB and IκB in cells with 20 nM IL-10 siRNA compared with the control.
