Effect of intracellular ThTP levels on AThTP accumulation. BL21 strains overexpressing E. coli AK (○) or GST-hThTPase (●) were grown overnight in LB medium containing ampicillin (0.1 mg/mL). The cultures were diluted to a density of A600 = 0.6 - 0.8 and protein expression was induced with IPTG (1 mM) for 3 h. Then the bacteria were transferred to a minimal medium containing 10 mM glucose without (A, C) or with CCCP 50 μM (B, D) and ThTP and AThTP were determined as a function of time. For comparison an experiment with the control BL21 strain (▲) is also shown. (Means ± SD, n = 3)
