Southern blot analysis. DNA isolated from the parental diploid strain (lane P), one of the Arg+ heterozygotes (isolate #19; lane H), and fourteen independent Arg+ Ura+ segregants were digested with Bgl II and resolved by agarose gel electrophoresis. A photograph of the ethidium bromide-stained gel is shown in panel C. The positions and sizes (kbp) of DNA size markers are indicated on the right. The DNA was transferred to a Hybond membrane, which was serially hybridized to 32P-labeled DNA probes A (panel A) and B (panel B) derived from the 5' and 3' segments of the CaCET1 gene, respectively.
